Abstract

This study investigated the relationship between dithiothreitol-oxidation potential (DTT-OP) and reactive oxygen species (ROS) depending on ratio of organic carbon (OC) and elemental carbon (EC) in PM2.5 from May 28 to June 18 of 2016 at the west coastal site in the Republic of Korea. Twenty-two integrated samples were collected for both DTT-OP and ROS analysis to investigate the main chemical classes of toxicity. The influence of atmospheric transport processes was assessed by the Residence-Time Weighted Back trajectory for the two selected sampling periods based on source region. The good correlation (r2 of 0.67) between of ratio of ROS to DTT-OP and ratio of OC and EC depending on source regions illustrate that primary long-range source including relatively higher EC was highly associated with higher ROS activity. This study provides insight into the sources in PM2.5 that can be drivers of the analytical approach for oxidative potential.

1. 서 론

대기 중 초미세먼지 (PM2.5)는 자동차 배출, 식생연소, 광화학반응에 의한 2차 성분, 그리고 1차, 2차 성분을 모두 포함하는 장거리 이동 성분으로 이루어져 있다 (Oh et al., 2018b, 2017; Park et al., 2016). 다양한 원인으로부터 발생된 초미세먼지는 최종적으로 기후변화 및 인체 유해성과 높은 관련성이 있다. 최근 연구에 따르면 초미세먼지에 대한 인체 노출은 호흡기 질환, 심혈관 질환, 신경발달 장애 등 건강에 직접적으로 악영향을 끼친다 (Cao et al., 2012; Sun et al., 2010). 수많은 유기성분 및 금속으로 이루어진 초미세먼지는 큰 표면적과 함께 폐조직 등으로 깊숙이 침투하여 산화 잠재력과 관련된 활성산소종 (reactive oxygen species, ROS)을 생성한다. 즉, ROS가 과하게 생성되면 염증, DNA 손상 및 세포사멸이 발생한다 (Xu et al., 2018; Vattanasit et al., 2014).

인체 유해성과 관련된 산화 잠재력을 분석하는 방법은 최근 여러 분석법이 개발되었다. 이 중 비세포 화학 반응방법인 디티오트레이톨 (dithiothreitol, DTT) 방법과, 세포 내 산화환원반응 상태를 기반으로 독성의 강도를 측정하는 ROS 방법이 많이 활용되고 있다 (Verma et al., 2012; Xia et al., 2004). DTT 법은 비세포 화학 반응으로 시료처리량과 경제적 측면에서 장점이 뛰어나며 (Ayres et al., 2008), 세포 내 산화스트레스, 병리학적 관련성이 있다 (Strak et al., 2017; Bates et al., 2015). 또한, DTT 방법은 세포 내 헴옥시게네이즈-1 (hemeoxygenase-1; HO-1)반응과 상관성이 있으며 (Li et al., 2003), 3-[4,5-dimethythia\-zol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT), tumor necro sis factor-α (TNF-α), interleukin-6 (IL-6) 같은 염증반응을 자극하는 사이토카인의 분비와 관련성이 있다 (Steenhof et al., 2011). 과거 연구를 살펴보면, 고기구이 초미세먼지 내 콜레스테롤 및 산화 잠재력과의 높은 상관성이 있으며 (Lee et al., 2018), 계절적 변화에 따른 PM2.5 농도와 DTT와 상관성 (Wang et al., 2019), DTT 산화환원 활성 강도와 미세먼지 내 유기탄소 (organic carbon, OC), 원소탄소 (elemental carbon, EC) 및 다환방향족탄화수소 (poly aromatic hydrocarbon, PAH)와의 관련성 (Hu et al., 2005), DTT 상대 고강도가 미세먼지 내 유기성분 및 금속성분 농도와의 높은 상관성 (Lin et al., 2019) 등 미세먼지 내 오염성분과 DTT와의 높은 상관성은 이미 많은 연구에서 보고되었다. 하지만, DTT 방법은 비세포방법으로서 실제 생물학적 반응을 나타내기에는 한계가 있으며 (Xiong et al., 2017; Landreman et al., 2008), 철과 같은 금속성분의 영향으로 DTT 결과에 대해 불확도가 증가할 수 있다 (Charrier et al., 2015; Ayres et al., 2008).

이에, 활성작용을 직접적으로 이해하기 위해 세포 내에서 생성하는 ROS를 이용하여 산화 잠재력을 분석한다. ROS는 세포분화, 유전자의 발현 등 다양한 세포 내 생물학적 과정에 연관되어 있기 때문에 (Rhee et al., 2000), ROS의 항상성을 유지하는 것은 세포 성장과 생존에 매우 중요하다. 하지만, 미세먼지 등에 의해 발현된 세포 내 산화스트레스는 ROS의 생성과 이를 제거하는 항산화 반응 간의 불균형으로 나타난다. 이로 인한 세포 내 ROS 증가는 DNA, 단백질, 지질과 직간접적으로 반응하여 세포를 손상시킨다. 더 나아가, 인체 노화나 심장과 관련된 질병들의 핵심 원인으로 알려져 있다 (Ozben et al., 2007). 과거 연구에 따르면, DTT 및 ROS는 계절별 배출되는 화학성분과 높은 상관성을 나타내며 (Ahlam et al., 2019; Hedayat et al., 2016), PM의 수용성 및 불수용성 성분이 세포독성에 중요한 역할을 한다 (Wang et al., 2013). 이는, 미세먼지 내 EC, OC, PAH 등을 포함한 유기지표성분, 미량 원소 성분과 산화환원 활성산소종 사이에 높은 상관관계가 있다 (Geller et al., 2006). 하지만 ROS는 분석과정 중 반응 중간 산물인 H2O2와 반응 가능성이 있기 때문에, 결과가 과대평가 될 수 있다 (Kalyanaraman et al., 2012; Dikalov et al., 2007).

본 연구에서는 안면도 기후변화감시소에서 포집한 시료에 대해 열광학 탄소분석기를 이용하여 OCEC 열광학 특성과 DTT 및 ROS를 이용한, 두 산화잠재력 간의 상관성을 분석하였다. 초미세먼지 내 유기탄소 및 원소탄소 농도 변이에 따른 DTT 및 ROS 분석을 통해 초미세먼지 성분에 따른 두 방법 간의 산화 잠재력 상관성을 분석하였다.


2. 연구 방법
2. 1 포집장소

분석시료는 충청남도 태안군 안면읍에 위치한 기후변화감시소 (36.32°N, 126.19°E, 해발 46 m)에서 고용량 포집장치 (Tisch Environmnetal, Inc., USA)를 이용하여, 석영필터 (2500QAT-UP, Pall Corp., USA)에 PM2.5를 2016년 5월 28일부터 6월 18일까지 총 22개 시료를 포집하여 OCEC, DTT-OP 및 ROS를 분석하였다.

2. 2 분석 방법

본 연구에서는 열광학 특성과 DTT 및 ROS를 이용한 두 산화 잠재력 상관성을 분석하고자, NIOSH5040 분석법을 기초로 한 탄소분석기 (Carbon Aerosol Analyzer, Sunset laboratory Inc., USA)를 이용하였다. 현장 공시료, 외부표준물질에 따른 내부표준물질의 정확성, 기기안전성, 재차분석 등을 통해서 최종 OCEC 농도의 신뢰성을 100±5% 이내로 유지하였다.

2. 3 디티오트레이톨 (dithiothreitol, DTT) 방법

산화환원반응인 DTT법은 선행연구에서 자세히 나타냈다 (Lee et al., 2018). 간략히, PM2.5 내 활성산소 성분과 DTT와의 반응, 잔류 DTT의 발색반응으로, 반응 시간에 따른 잔류 DTT와 반응 생성물인 5-mercapto-2-nitrobenzoic acid (thionitrobenzoate, TNB)를 흡광 (412 nm) 측정하여 활성산소의 농도를 산출한다. 본 연구에서는 2 mM DTT, 2 mM DTNB와 함께, 100 mM potassium phosphate dibasic, potassium phosphate monobasic, 1% trichloroacetic acid를 이용하였다. 전체 10 mL 혼합액이 37°C 등온 교반 하에, 반응 시작 후 40분까지 총 5회 흡광도 412 nm에서 TNB를 분석하였다. 최종 DTT 소비 감소율 (DTT consumption rate (nmol/min))은 반응 시간 내 5회 분석된 흡광도의 감소 기울기 (σABS)에 의해 바탕 시료의 흡광도 감소 기울기를 보정한 후, 포집 부피 농도로 정규하여, 식 (1)과 같이 dithiothreitol-oxidative potential (DTT-OP)를 산출하였다 (Lee et al., 2018).

본 연구에서 시료 흡광 감소 기울기의 상관관계 결정계수의 평균은 0.996로 매우 높게 나타났다.

2. 4 활성산소종 (reactive oxygen species, ROS) 2,7-dichlorodihydrofluorescein diacetate (DCFH-DA) 방법 (ROS-DCFH-DA)

본 연구에서는 ROS를 분석하기 위한 방법으로 2,7-dichlorodihydrofluorescein diacetate (DCFH-DA)를 활용하였다 (그림 1). DCFH-DA 방법은 일반적으로 hydroxyl radical, peroxide, superoxide radical, peroxynitrite 등과 같은 활성산소 성분과 반응한다 (Landreman et al., 2008). 초미세먼지에 대한 활성강도를 측정하기 위해, 배양된 쥐의 폐대식세포 (NR8383, American Type Culture Collection)를 이용하였다. 대식세포에 비형광 반응물질인 2,7-dichlorodihydrofluorescein diacetate (DCFH-DA)를 주입하면, 세포 내 빠른 디아세틸화 반응에 의해 dichlorodihydrofluorescein (DCFH)로 변환된다. 이후, 초미세먼지의 자극에 의한 ROS 생성이 형광 최종 산물인 2,7-dichlorofluorescein (DCF)을 생성한다. 폐대식세포에 대한 초미세먼지는 37°C, 6% CO2 대기 조건에서, 2.5시간 동안 노출시켰으며, 504 nm (Excitation), 529 nm (Emission) 하에 분석되었다. 최종 분석자료는 Zymosan positive control로 정규화하여 정량을 산출했다 (Landreman et al., 2008).

3. 결 과

선행연구 (Oh et al., 2018a)에서 기상청 기후변화감시소에서 2016년 5월 28일부터 22일간 포집된 시료를 각각 3가지 다른 탄소분석 프로토콜 (NIOSH5040, IMPROVE_A 및 EUSAAR2)에 의해 TC, OC, EC 분석한 결과, TC의 경우 세 가지 분석법 모두 통계적 유의 수준에서 모두 같게 분석되었으나, NIOSH5040와 IMPROVE_A에서 통계적으로 OC 및 EC의 분율이 다르게 나타났다. NIOSH5040 프로토콜은 OC 분석단계에서 최대 870°C로, IMPROVE_A 분석법 580°C에 비해 상대적으로 높은 온도를 사용하며, 광투과 및 광반사법에 의해 OCEC를 분리한다. 본 연구에서는 NIOSH5040 프로토콜에 의한 OCEC를 결과를 활용했다.

본 연구기간에 분석된 OC의 경우 2.19±1.46 μg/m3 (평균±표준편차), EC 0.22±0.06 μg/m3으로 나타났으며, OC와 EC의 비율 (OC/EC)은 8.60±3.05로 나타났다 (표 1). Zymosan Unit (ZU)로 정규한 ROS는 최소 49에서 최대 475 사이에서 193±118 μg/m3으로 분석되었으며, DTT-OP는 1.90±0.31 nmol/min/m3로 분석되었다. ROS/DTT-OP은 100±53으로 나타났다. 기존 연구에서 보고된 DTT 농도를 살펴보면, 미국 로스엔젤레스 및 중국 베이징에서 봄에 측정된 농도는 각각 약 0.21 nmol/min/m3 (Hu et al., 2008), 0.24 nmol/min/m3 (Liu et al., 2014)로 나타났고, 미국의 여름, 가을, 겨울에 측정된 농도는 약 0.3, 0.34, 0.33 nmol/min/m3 (Verma et al., 2014)로 나타나, 본 연구 결과에 비해서 낮게 나타났다. 이는 DTT 반응 시약의 첨가 비율 등 분석 조건이 상이하여, 본 연구 결과와 흡광 감소 기울기를 상대 비교할 수 없다. 향후, DTT를 9,10-Phenanthrenequinone (CAS# 84-11-7) 등과 같은 quinone 성분의 흡광 감소 기울기를 기준으로 정규화한다면 분석 결과에 대한 상대 비교 평가를 할 수 있다.

반면에 ROS의 경우 Zymosan positive control로 정규화하여 정량을 한 경우 상대 비교 평가가 가능하다. 그리스 Athens에서 여름 약 150 μg/m3, 겨울 약 80 μg/m3 (Taghvaee et al., 2019)로 나타났다. 레바논 Beirut의 고속도로 근처와 도시에서 645.6 μg/m3, 약 420 μg/m3 (Daher et al., 2014), 이탈리아 Milan에서 1월, 2월, 5월, 6월 각각의 농도는 약 837, 920, 193, 240 μg/m3 (Daher et al., 2012)로 나타나 본 연구결과에 비해서 다소 높게 나타났다. 이는 본 측정 장소인 기후변화감시소가 안면도 교외지역에 위치하여 상대적으로 낮은 값이 나타났다.

본 연구기간 동안 분석 농도 시계열을 보면 (그림 2), ROS는 분석 초기 5월 28일 최대 475 μg/m3와 6월 1일 469 μg/m3로 두 측정일의 비율이 약 1.01로 거의 유사한 반면, 동일 측정일에 대한 DTT-OP 농도 비율은 1.46으로 5월 28일 농도가 크게 분석되었다. 이와 유사하게, 6월 6일 각각의 두 ROS, DTT-OP 상대 농도 값도 차이를 나타냈다. 더 나아가, OC 및 EC 농도 값을 살펴보면, OC의 경우 6월 1일 5.5 μg/m3, 6월 6일 2.3 μg/m3으로 두 측정일의 비율이 약 2.4인 반면, EC의 경우 1.7로, 6월 1일 OC의 농도가 EC에 비해 상대적으로 높은 값을 나타냈다. 6월 1일의 OC/EC는 13.9, ROS/DTT-OP는 231, 6월 6일은 각각 9.8, 89.2로 분석되었다. 이는 OC/EC와 ROS/DTT-OP가 상관성이 있음을 나타낸다.

이를 좀 더 자세히 분석하고자, 그림 3에 ROS/DTT-OP와 OC/EC의 상관관계를 측정일에 대해 칼라 표기하여 나타내었다. 상관관계분석 결과, 기울기 14.13에서 상관관계결정계수 (r2)가 0.67로 높게 나타났다. 이는 앞서 분석한 두 활성산소 분석법이 초미세먼지 내 탄소 성분, 특히 OC 및 EC의 비율과 관련성이 있다. 즉, 상대적으로 높은 EC 농도에서 높은 ROS, 또는 낮은 DTT-OP 값을 나타냈다. 이는 초미세먼지를 구성하는 성분, 더 나아서 발생 원인이 DTT-OP 및 ROS 사이의 두 분석법에 영향을 줄 수 있다. 비세포 DTT 방법의 경우, 앞서 설명한 바와 같이 금속성분이 DTT 발색에 간섭을 줄 가능성이 있다 (Charrier et al., 2015). 이와 더불어, 본 연구결과에서는 (1) 상대적으로 높은 농도의 EC가 존재하는 경우, 또는 (2) EC 및 중금속을 포함한 1차 연소 배출성분이 초미세먼지 내 상대적으로 높은 농도로 존재할 경우, DTT 분석에 과대평가가 될 수 있다. (3) 또한, 상대적으로 높은 OC가 존재하는 경우, 높은 ROS가 분석되었다.

그림 2의 시계열 농도를 살펴보면, 6월 6일을 기점으로 상대적인 농도가 줄어드는 것으로 분석되었다. 이에 두 기간 (5월 28일~6월 6일, 6월 7일~6월 19일)으로 나누어 농도를 살펴보면, OC의 경우 3.33 μg/m3, 1.31 μg/m3로 첫 번째 기간이 약 2.5배 높게 나타났다. 또한, EC는 1.5배, ROS는 2.1배로 높게 분석되었다. 이에 고농도 기간 동안 발생 지역의 오염원을 분석하고자 오염원별 역궤적 체류 가중치 농도 모델 (Residence-Time Weighted Back trajectory, RTWB)를 수행하였다. 본 모델은 각 격자안 누적 역궤적 기류를 분석하여 각 격자 누적수에 대한 ROS/DTT-OP 가중치를 적용한 결과를 그림 4에 나타냈다.

첫 번째 기간 동안 RTWB 결과를 보면, 중국 칭다오 시 주변 기류가 영향을 미친 것으로 나타났으며, 두 번째 기간 동안에는 서해 일부 및 우리나라 서해안 지역에서 발생된 초미세먼지가 측정소에 영향을 주었다. 즉, 중국발 미세먼지가 장거리 이동 시 EC를 포함한 1차 오염원이 상대적으로 높게 나타나며, 이는 ROS에 상대적으로 크게 영향을 주었다. 서해안 발생 초미세먼지는 상대적으로 2차 성분이 크게 나타날 가능성이 있으며, ROS, DTT-OP 모두에 영향을 주는 것으로 해석된다. 이를 좀 더 분석하고자 탄소 분석기로부터 원자료를 추출하여 기간별 열광학 분포를 분석하였다.

본 연구에서 분석된 탄소 분진 특성을 세밀히 분석하고자 그림 5에 열광학 분포를 나타냈다. 탄소 열광학 분포는 높은 휘발성 탄소 성분은 저온에서 분석되며, 결합력이 강한 성분은 고온에서 분해되기 때문에, 시료 내 탄소 성분의 특성을 이해할 수 있다. 탄소 분석기로부터 분석된 원자료에 대해서 시스템 바탕 값 보정, 광투과율 등을 확인 후, 동시간자료 TC 농도 값으로 정규화하여 열광학 분포를 최종 계산하였다. 그림 5(b)의 y축은 분석 날짜 별 분석 시간 대응 승온 단계를 나타낸다. 적색 면적은 TC 농도로 정규화 (dCH4 ppm dt-1/TC μg/cm2 dt-1)한 탄소 열광학 분포이다.

그림 5(a)에 나타내듯이 첫 번째 기간 열광학 분포에서, 산소 (O2 in He) 주입 분석 조건에서 OC로 분석되는 Pyrolytic carbon (PC)이 두 번째 기간보다 높게 나타났다. 두 번째 기간 열광학 분포에서는 첫 번째 기간에 비해 낮은 PC 및 He 분석 상태에서 고온 분석된 OC가 높은 것으로 나타났다. 최근 연구결과에 따르면, PC는 식생연소에 따른 Brown Carbon (Brc)과 연관성이 있다 (Massabò et al., 2016). 이는 장거리 이동 시 EC를 포함한 1차 오염원은 대기 중 BrC를 가중시키며, ROS에 DTT보다 큰 영향을 줄 수 있을 것으로 사료된다.

본 연구 결과는 대기 중 미세먼지 내 산화독성을 이해하기 위해서, 향후 연구자들이 분석방법을 선택하는 기준을 제공하는 데 의의가 있다. 더 나아가, 우리나라는 무기탄소를 포함하는 황사 등 토양 기원 원인이 다른 나라에 비해 상대적으로 크게 나타나며, 2차성분이 크게 차지함을 고려해 볼 때, 이를 분석할 수 있는 산화 독성법을 기준으로 선택하는 것이 향후 일관성 있는 자료 해석에 많은 기여를 할 수 있다.


4. 결 론

본 연구의 분석시료는 기후변화감시소에서 PM2.5를 2016년 5월 28일부터 6월 18일까지 총 22개 시료를 포집하여 열광학 탄소분석기를 이용하여 OCEC 열광학 특성과 DTT 및 ROS를 이용한, 두 산화잠재력 간의 상관성을 분석하였다. 결과, 중국발 미세먼지가 장거리 이동시 EC를 포함한 1차 오염원이 상대적으로 높게 나타나는 경우, DTT 분석에 과대평가가 될 수 있으며, 상대적으로 높은 OC가 존재하는 경우 높은 ROS가 분석되었다. 더 나아가, 식생연소 등에 의한 장거리 1차 오염원은 대기 중 EC 및 BrC를 가중시키며, ROS에 DTT보다 큰 영향을 줄 수 있을 것으로 사료된다. 본 연구결과는 향후 국내 미세먼지 독성방법 및 자료 해석에 기본적인 방향 제시를 할 수 있을 것으로 판단한다.
